Dr. Paul Lombroso, YaleA Potential Therapy for Fragile X Moves Forward

An exciting new collaboration is bringing a potential drug for Fragile X closer to reality! Yale University professor Dr. Paul Lombroso and the pharma company GlaxoSmithKline have joined forces to further develop drugs targeting STEP (striatal-enriched protein tyrosine phosphatase) in Alzheimer’s disease.

With FRAXA funding from 2008 through 2011, ;Dr. Lombroso's team showed that STEP also plays a critical role in Fragile X. Overactivity of STEP in Fragile X syndrome results in abnormal synaptic plasticity, and reducing STEP in Fragile X mice reverses a wide range of symptoms.

Dr. Lombroso and his team are now working to find viable drugs to reduce STEP activity and thus treat Fragile X and other diseases with STEP dysregulation and cognitive deficits, such as Alzheimer’s and schizophrenia. Their partnership with GlaxoSmithKline should speed up development of this new class of drugs which have real potential for treating Fragile X.
